控制蟠桃果实扁平性状的候选基因分析

摘 要:

蟠桃(Prunus persica ‘Compressa’)果实扁平性状是质量性状, S (saucer-shaped)位点控制。利用番茄(Lycopersicon esculentum)果实形状控制基因, 搜寻蟠桃基因组中S位点的候选基因, 筛选到一个基因——Ovate Family Protein 1 (PpOFP1); 利用转录组数据, 分析候选基因在不同组织中的表达模式, 确定其编码一个由421个氨基酸构成的卵形家族蛋白(OVATE family protein), 含有N端核定位信号和Covate结构域, 为拟南芥(Arabidopsis thaliana) AtOFP1直系同源物。利用实时荧光定量PCR (qRT-PCR)方法检测候选基因在不同桃树品种中的表达模式, 发现PpOFP1基因在被检测的蟠桃品种中的表达量均显著高于圆桃品种以及蟠桃芽变后的圆形桃。综上, 桃树PpOFP1是一个控制蟠桃果实扁平性状的重要基因, S位点的候选基因。

Abstract:

The flat-shape trait of peach (Prunus persica) Compressais known to be controlled by a major S (saucer-shaped) gene locus. In this study we try to determine the candidate genes for the S locus. Based on the cloned genes that determine the shape of tomato fruit, we tried to identify and fnally fgured out that ovoid gene PpOFP1 might be candidate gene of S locus. Public RNA-seq data were used to analyze the expression pattern of PpOFP1 in various tissues, and to manually determine the open read frame of PpOFP1. The results show that it codes a 421 aa poly-peptide, and the N terminal contains a nuclear localizing signal, while the C terminal includes a ovate domain. The analysis of homology by using InParanoid and MCScanX softwares show that ovate gene AtOFP1 in Arabidopsis thaliana was homologous to the candidate gene PpOFP1 in peach. The qRT-PCR experiment on different peach varieties showed that the transcripts of PpOFP1 in flesh of all ‘Compressa’ varieties tested were higher than those of round peach varieties, and the transcripts of PpOFP1 in ‘Compressa’ were higher than its bud sport whose fruit shape was round. In conclusion, PpOFP1 is a key gene that controls the flat-shape trait of ‘Compressa’, and it is a likely candidate gene for S locus.
